The use of additive-manufactured components in medical applications, specifically medical devices (e.g., orthopedic casts), has increased in recent years. Such devices may be fabricated at the point of care using consumer-grade additive manufacturing Limited studies have been conducted to evaluate the extractable substances of these devices. Chem. characterization followed by toxicol. risk assessment is one means of evaluating safety of devices. This study was designed to determine the extractables profile of additive-manufactured materials according to filament grade and post-processing method. Feedstocks for additive manufacturing were tested as filament and manufactured casts, while the cast from consumer-grade filament (CGF) was post-processed. Samples were extracted using three solvents of varying polarities. Extracts were analyzed by gas chromatog./mass spectrometry (GC/MS) and liquid chromatog./mass spectrometry (LC/MS) techniques. In GC/MS anal., isopropanol extracts generated fewer compound identifications for USP Class VI filament (USPF)-based casts (3) compared with the resp. filament (17) while hexane generated the most compound identifications for the finished cast manufactured from CGF. CGF was found to have the highest number of nonvolatile extractables for isopropanol (15) and hexane (34) by pos. ion LC/MS. Addnl., CGF produced more non-polar extractables in hexane than the USPF. A known polymer byproduct and potential genotoxicant, styrene acrylonitrile (SAN) trimer, was one of the compounds identified in both GC/MS and LC/MS at quantities ranging from 19 to 270μg g-1. Overall these results suggested that the extractables profile was affected by the filament material, printing procedure, and post-processing method.

An extensive literature search revealed that there exist very few methods on RP-HPLC reported and HPTLC method was not reported. This study deals with simple, precise, accurate and economical stability indicating HPTLC method development with validation as per ICH guidelines with application of developed method on in Apremilast pharmaceutical formulations and Identified and Characterization of degradation products using MS/MS. Method: Precoated Silica gels plates were used as stationary phase. Toluene: Et Acetate (4:6; volume/volume) was delivered best separation at 236 nm (Rf 0.55 ± 0.02) by densitometry anal. Degradation anal. was performed as per ICH guidelines (Q2R1). Isolation of degradation product by HPTLC method and identify by MS/MS method. Results: The linearity was 100-600 ng with R2 of 0.997 while, % RSD was in range. LOD and LOQ of Apremilast were found 0.77ng/spot and 2.35ng/spot resp. The recovery of Apremilast was found to be 99.70 ± 0.23%. The % assay of active substance was found in a range 80.62 to 100. HPTLC and MS/MS method revealed possible degradation mechanism of 11 degradant products. Conclusion: The Proposed developed and validated HPTLC method was found to be more sensitive, simple, precise, accurate, cost effective and robust. This method could be applied for anal. of bulk drug and tablet formulation, degradation study. This degradation pathway of drug will help to identify the degradation products of Apremilast.

Besides, there were two unknown impurities like 3-(amino-1,3-dioxoisoindolin-2-yl)phthalic acid and 4-aminoisobenzofuran-1,3-dione identified by HPLC. A thorough study was under taken to synthesize and characterize these unknown impurities. This improved process has a number of industrial advantages, such as economical material cost and relatively high yield. The structure of the synthesized compounds was elucidated by IR, 1H NMR, 13C NMR, mass spectroscopy and elemental analyses.

This study aims at investigating the separation, identification and characterization of related substances in apremilast by LC-MS hyphenated techniques, as well as the synthesis optimization and the degradation pathways elucidation. Forced degradation studies were conducted under the ICH prescribed stress conditions. The chromatog. separation was achieved on XBridge C18 column (4.6 mm × 150 mm, 3.5 μm) using a mobile phase consisting of water adjusted to pH 3.0 with formic acid as solvent A and acetonitrile as solvent B in linear gradient elution program. Twelve related substances were detected all together in apremilast and its stress samples. Their structures were identified mainly through pos. ESI high-resolution TOF-MS anal. of the parent ions’ accurate masses and elemental compositions, and the corresponding MS/MS spectra elucidation. There were three process-related substances and nine degradation products, seven of them were first reported. Two degradation products and one process-related substance were further verified by semi-preparation and NMR determination Their origins and formation mechanisms were also discussed, based on which effective approaches for the synthesis optimization were conducted. Therefore, the related substances investigation are valuable for apremilast manufacturing process optimization and quality control.

The aims of this study were to analyze the metabolite profiling results of various extract of Chyrsophyllum cainito L. leaves, which contain phytoestrogen, through in silico study against 3OLS protein, an X-ray protein of ERβ, so it can predict the types of the phytoestrogen contents which have antiosteoporosis property. In silico anal. was carried out for the compounds from the metabolite profiling data of C. cainito leaves from our previous study. The structure compounds from metabolite profiling results of various extract of C. cainito leaves were prepared with Avogadro 1.0.1 software, mol. docking was done using PyRx 0.8 software, and Biovia Discovery Studio Visualizer 2016 software was used to visualize the structure of compounds against 3OLS protein. The physicochem. characteristics of the compounds were analyzed using the SwissADME web tool. It was known that there were total 11 compounds in C. cainito leaves that predicted as phytoestrogens which have ERβ agonist properties against 3OLS protein. The ERβ agonist was a compound that has parameters similar to 17β-estradiol in its interaction with 3OLS protein, which has a pharmacophore distance of 10.862 Å, and binding to amino acids His 475 and Glu 305 or Arg 346 at receptor-ligand docking simulation. C. cainito leaves contain 11 compounds that are predicted to be phytoestrogens with ERβ agonist properties, which is responsible for antiosteoporosis activity.
