A mixture of methyl 2-methyl-5-nitrobenzoate (1.0 g, 5.12 mmol) and l-ie/t-butoxy- N,N,N’,N’-tetramethylmethanediamine (2.23 g, 12.81 mmol) was heated at 1 15 C for 2 h. Then the reaction mixtuer was concentrated and purified by column chromatography to afford 600 mg of (E)-methyl 2-(2-(dimethylamino)vinyl)-5- nitrobenzoate. To a solution of (E)-methyl 2-(2-(dimethylamino)vinyl)-5- nitrobenzoate (600 mg, 2.40 mmol) in toluene (5 mL) was added (2,4- dimethoxyphenyl)methanamine (602 mg, 3.60 mmol) and the reaction mixture was heated at 120 C for 2 h. Then the reaction mixture was concentrated and the residue was triturated with EtOAc: Et20 (1 :9) and the solid precipitate was filtered and dried to afford 700 mg of the title product. 1H NMR (400 MHz, DMSO- 6): delta 9.32-9.31 (d, = 2.4 Hz, 1H), 8.41-8.38 (dd, = 2.4, 8.8 Hz, 1H), 7.62-7.60 (d, = 9.6 Hz, 1H), 7.48-7.46 (d, J = 7.2 Hz, 1H), 7.44-7.42 (d, J = 8.0 Hz, 1H), 6.52-6.49 (m, 3H), 5.15 (s, 2H), 3.86 (s, 3H), 3.82 (s, 3H).

The organic phase was then dried over Na2SO4 and evaporated to give a crude solid product 59 that crystallized from ethyl ether. Filtration and open air suction drying afforded an off-white solid pure product 9.8 g (96%), (MS: [M+1]=347).

Heat the mixture to 95-100 C and stir at that temperature for 16 hours. Cool to 20-25 C and collect the precipitated solid by filtration. Dry the yellow solid in the air for two days to give the crude product (2.17 Kg, 66%) which is used without further purification. ES/MS (m/z): 341.1 (M+H).

1-(2,4-Dimethoxyphenyl)methanamine (20.3 mL, 135 mmol) was added to asolution of C4 (10.3 g, 90.2 mmol) in methanol (200 mL), and the reaction mixture was stirred for 1 hour at room temperature. It was then cooled to -78 C; lithium borohydride solution (2 M in tetrahydrofuran, 45.1 mL, 90.2 mmol) was added drop-wise, and stirring was continued at -78 C for 2 hours. After slowly warming to room temperature overnight, the reaction mixture was quenched via careful addition of saturated aqueous sodium bicarbonate solution. Ethyl acetate (250 mL) and sufficient water to solubilize the precipitate were added, and the aqueous layer was extracted with ethyl acetate; the combined organic layers were dried over magnesium sulfate, filtered, and concentratedin vacuo. Silica gel chromatography (Gradient: 0% to 5% methanol in dichloromethane)provided the product as a colorless oil (10.4 g). Similar purification of mixed fractionsafforded additional product (3.7 g). Combined yield: 14.1 g, 53.1 mmol, 59%.
