Rasagiline mesylate (RSM) is a selective and irreversible monoamine oxidase B inhibitor used for the treatment of Parkinson’s disease (PD). However, its unfavorable biopharmaceutical properties, such as extensive degradation in the gastrointestinal tract and first-pass metabolism are responsible for its low oral bioavailability and suboptimal therapeutic efficacy. Here, we report the feasibility of delivering RSM via the transdermal route using RSM containing microemulsion-based gel (RSM-MEG) to achieve effective management of PD. Our in vitro skin permeation studies of RSM-MEG showed significantly higher (at least ∼1.5-fold) permeation across rat skin compared to the conventional RSM hydrogel. Our skin irritation studies in rabbits showed that RSM-MEG is safe for transdermal application. Finally, using the rat model of rotenone-induced Parkinsonism, we demonstrated that the topical application of RSM-MEG was equally effective in reversing PD symptoms when compared to oral RSM therapy. Thus, our study confirmed the feasibility and potential of transdermal delivery of RSM via simple topical application of RSM-MEG, and this approach could be an alternative therapeutic intervention for the treatment of Parkinson’s disease.

Principal component anal. was used to compare the gas chromatograph high-resolution time-of-flight mass spectrometry data for differences/similarities in their clustering. When egg whites were substituted with 30% mushrooms, the total volatile abundance increased 8-11 times, compared to egg white alone. The dominant volatiles in the egg white-mushroom blends were aldehydes and alcs. followed by ketones, heterocyclic compounds, and sulfurs. Increases in the mushroom proportions led to a significantly higher peak abundance of all volatiles in the blends except sulfurs. Crimini imparted higher amounts of heterocyclic compounds, and white mushrooms had higher sulfurs. Compared to the steam method, roasting significantly increased pyrazines in the blends. This study demonstrated the contribution of mushroom volatiles to egg whites, three impact factors, and the potential utilization of mushroom in food product development.

Previous studies on the application of a novel multifrequency ultrasound washing treatment have proven to be effective in guaranteeing microbial safety, and enhancing the quality of cherry tomato. Nevertheless, there is still a need to evaluate the effect of these treatments on the metabolic activity, volatile profile, and sensorial quality of the fruit. This study evaluated the above parameters of cherry tomato treated with optimized assisted and un-assisted multifrequency ultrasound. The treatments slightly enhanced the enzyme activities and inhibit ethylene production and respiration rate. The results indicated that the treatment with 20/40 kHz and aqueous ozone (0.85 ± 0.2 mg/L) for 10 min enhanced and maintained the volatile compounds (VC). Out of the 44 identified VC, one had odor activity value > 1 and thereby considered as active aroma compound contributing significantly to the aroma of the sample. E-nose anal. indicated that alc., terpenes, and some aromatic compounds are the core indicators of the significant sensors. PCA showed that the processing methods had an influence on the flavor indexes of the tomatoes. The result verified that all the washing techniques did not result in a substantial neg. effect on the tomato, and the treatment with ultrasound and ozone proved to be the best.

This study monitors the presence of 88 volatile organic compounds (VOCs) and semi-volatile organic compounds (semi-VOCs) at the gas phase of seven indoor settings in a school in the city of Tarragona, Spain, and five outdoor locations around the city. The VOCs and semi-VOCs monitored were solvents (∑Solvents), aldehydes (∑Aldehydes), emerging organic compounds (∑EOCs), and other VOCs and semi-VOCs (∑Others). Passive sampling campaigns were performed using Carbopack X tubes followed by thermal desorption coupled to gas chromatog. with mass spectrometry (TD-GC-MS). Overall, 70 of the target compounds included in the method were determined in the indoor air samples analyzed, and 42 VOCs and semi-VOCs in the outdoor air samples. Our results showed that solvents were ubiquitous throughout the school at concentrations ranging from 272μg m-3 to 423μg m-3 and representing 68%-83% of total target compounds (∑Total). The values of ∑Total in 2021 were three times as high as those observed at the same indoor settings in 2019, with solvents experiencing the greatest increase. A plausible explanation for these observations is the implementation of anti-COVID-19 measures in the indoor settings, such as the intensification of cleaning activities and the use of hydroalcoholic gels as personal hygiene. The ∑Total values observed in the indoor settings evaluated were twenty times higher than those found outdoors. ∑Solvents were the most representative compounds found indoors (74% of the ∑Total). The concentrations of VOCs and semi-VOCs observed in the outdoors were strictly related to combustion processes from automobile traffic and industrial activities, with ∑Others contributing 58%, ∑Solvents 31%, and ∑Aldehydes 11% of the ∑Total. EOCs, on the other hand, were not detected in any outdoor sample.

Nanostructured lipid carrier (NLC) formulations were prepared using optimized ART concentration of 0.25 g dissolved in 5% w/v mixture of solid (Gelucire 43/01 and Phospholipon 85G) and liquid (Transcutol) lipids at 90°C. An aqueous surfactant phase at 90°C was added (dropwise) under magnetic stirring (1000 rpm) for 5 min. The pre-emulsion was speedily homogenized at 28,000 rpm for 15 min and further probe sonicated at 60% amplitude (15 min). Resultant sample was cooled at room temperature and frozen at -80°C prior to lyophilization. The freeze-dried sample was used for solid-state characterization as well as in the formulation of transdermal nanogels using three polymers (Carbopol 971P, Poloxamer 407, and Prosopis africana peel powder) to embed the ART-NLC, using ethanol as a penetration enhancer. Transdermal ART-nanogels were characterized accordingly (phys. examination, pH, drug content, rheol., spreadability, stability, particle size and morphol., skin irritation, in vitro and ex vivo skin permeation, and anal. of permeation data), P < 0.05. Overall, ART nanogels prepared from Poloxamer 407 showed the most desirable drug permeation, pH, swellability, spreadability, viscosity, and transdermal antiplasmodial properties superior to PAPP-ANG > C971P-ANG. A two-patch/wk concurrent application of the studied nanogels could ofer 100% cure of malaria as a lower-dose (50 mg ART) patient-friendly regimen devoid of the drug’s many side efects.

N-acylhomoserine lactone (AHL)-based bacterial communication through quorum sensing (QS) is one of the main causes of biofouling. Although quorum quenching (QQ) has proven to be an effective strategy against biofouling in membrane bioreactors (MBRs) for municipal wastewater treatment, its applicability for industrial wastewater treatment has rarely been studied. This is the first study to isolate QQ strains from the activated sludge used to treat industrial wastewater containing toxic tetramethylammonium hydroxide (TMAH) and 1-methyl-2-pyrrolidinone. The two QQ strains from genus Bacillus (SDC-U1 and SDC-A8) survived and effectively degraded QS signals in the presence of TMAH. They also showed resistance to toxic byproducts of TMAH degradation such as ammonium and formaldehyde. They effectively reduced the biofilm formation of Pseudomonas aeruginosa PAO1 and mixed community of activated sludge. The strains isolated in this study thus have the potential to be employed to reduce membrane biofouling in MBRs during the treatment of TMAH-containing wastewater.
