Efinaconazole is the first azole derivative approved by FDA for the topical treatment of onychomycosis. The objective of present study was to develop and validate HPLC method for estimation of efinaconazole in ex vivo human nail permeation study samples. The chromatog. anal. was performed on a HPLC system equipped with diode array detector. The efinaconazole and internal standard (IS) were extracted from the human nail samples by using the protein precipitation method. The samples were injected on to 5μm Polar C18 100Å, 4.6 mm x 150 mm column. The mobile phase consisted of 0.01 M potassium dihydrogen phosphate: acetonitrile (36:64) and eluent was monitored at 205 nm. The chromatog. separation of drug and analyte was achieved using isocratic elution at flow rate of 1 mL/min with a total run time of 15 min. The efinaconazole and IS were eluted at 6.4 ± 0.5 and 8.3 ± 0.5 min, resp. The developed method was validated as per FDA guidelines, and the results met with acceptance criteria. The method developed was specific, and the analyte concentrations were linear at range of 50 to 10000 ng/mL (R2 â‰?0.9981). The validated HPLC method was applied for quantifying efinaconazole in human nail permeation study samples. The permeation of efinaconazole was increased by twofolds with Labarfac CC (15135.4 ± 2233.9 ng/cm2) compared to formulations containing Transcutol P (6892.0 ± 557.6 ng/cm2) and Labrasol (7266.1 ± 790.6 ng/cm2). The study results demonstrate that developed efinaconazole HPLC method can be employed for formulation evaluation and clin. studies.

Fisetin is a polyphenolic flavonoid reported to have antioxidant, anti-inflammatory, and anti-cancer activities. However, it loses its importance as an effective phytochem. due to its poor water solubility and lower bioavailability. In the present study, the self-nanoemulsifying drug delivery system (SNEDDS) of fisetin was developed in order to improve its pharmacol. activity. The developed SNEDDS of fisetin was evaluated for improving the rotenone-induced behavioral changes in the rats, and its efficacy was compared with naive fisetin. It was noticed that fisetin loaded in the SNEDDS formulation significantly (p < 0.001) ameliorated the rotenone-induced alteration in the body weight, grip strength, beam walk, postural instability, etc., in rats when compared to the effect of naive fisetin. Naive fisetin significantly (p < 0.05) ameliorated the effect of rotenone on the level of dopamine only at a higher dose. Whereas, SNEDDS of fisetin produced a significant (p < 0.05) effect at both dose levels when compared with the diseased group as well as also produced a significant (p < 0.05) effect when compared with the naive fisetin group. The results of histopathol. examination revealed about the neuroprotective effect of SNEDDS loaded with fisetin as observed through the protection of neuronal damage. From this study, it was concluded that SNEDDS improved the anti-Parkinsonian activity of fisetin by improving the behavioral alteration produced by rotenone due to enhancement in its solubility and bioavailability.

Colloidal crystals with non-close-packed structures, such as body-centered cubic (bcc) structures, are of high interest. Nanosized protrusions, prepared from colloidal crystals, are interesting for biomimetic applications. Herein, self-assembly approaches for synthesizing colloidal crystals with square lattice nanoprotrusions are presented. In the first approach, spin-coating and subsequent heating of colloidal silica and acrylic monomer results in a bcc-like arrangement; the number of stacked colloid layers and the colloid d. of these compounds are important in the formation of this structure. Moreover, prolonged heating exposes colloids from acrylic monomer, with square lattice nanoprotrusions formed. In the second approach, the protrusions are fabricated in a shorter time by replacing some of the acrylic monomer with volatile media. The objective of this research was to design a new microemulsion gel for transdermal delivery of Nifedipine. For its good solubilizing capacity and skin penetration capability, a microemulsion-based system was selected. Castor oil, Tween 20, and PEG 400 were screened as oil, surfactant, and cosurfactant resp. from the solubility study. From pseudo ternary phase diagrams, the concentration of oil phase, Smix, and distilled water were determined and further processed for the formulation of Nifedipine microemulsion. Nifedipine loaded microemulsion optimized by stability study and in vitro permeation study. 1 percent weight/weight Carbopol was used to develop a microemulsion gel formulation. Using Keshary Chien diffusion cells, the efficacy of several microemulsion formulations to transport Nifedipine through the rat skin was studied in vitro. The in vitro permeation data showed that microemulsion gel increased the permeation rate (0.373 mg/cm2/h) of Nifedipine compared with the control solution (0.129 mg/cm2/h). All microemulsion formulations had droplet sizes ranging from 69 to 82 nm. The studied microemulsion gels were stable toward all stability conditions. A skin irritation study suggested that Nifedipine loaded microemulsion and microemulsion gel formulation were nonirritating and did not cause erythema. Thus, microemulsion gel systems are a promising formulation for the transdermal delivery of Nifedipine.

Flutamide is a non-steroidal anti-androgenic agent which is not only used in treating prostate cancer but also can be effective in some disorders such as androgenic alopecia and male pattern baldness. Several serious side effects for systemic administration of flutamide can be overcome by emulgel dosage form. Absorption enhancers can promote permeation of flutamide through skin. Percutaneous absorption of Flutamide emulgels with different concentrations of Transcutol P and Precirol ATO5 was studied. Various emulgel formulations using different concentrations of Transcutol P and Precirol ATO5 with 0.5 to 5% weight/weight were prepared Percutaneous absorption was tested with standard Franz Diffusion Cell equipment using full thickness rat skin. Drug concentrations in the samples were analyzed by High-Performance Liquid Chromatog. (HPLC) equipped with UV-Vis detector at 305 nm. The percutaneous absorption of flutamide emulgels formulated with enhancers was higher than control formulations. The enhancement ratio increased from 1.218 to 3.670 and 1.346 to 3.900 for Precirol ATO5 and Transcutol P, resp. The area under the Curve (AUC) increased by increasing the enhancers’ concentration and a significant upsurge was seen in the concentration 1% for both enhancers. The flutamide emulgel containing 1% Transcutol P showed more appropriate percutaneous absorption through the skin compared to others.

Bisphosphonates are very poorly absorbed from the GI tract and have limited bioavailability of less than 1% with adverse gastrointestinal effects in humans. The present work aimed to develop sublingual spray formulation using Risedronate sodium with combination of Pullulan gum and HPMC E5. Formulation of sublingual spray was developed by employing optimization technique. The optimized formulation was selected and evaluated for various pharmaco-tech. parameters such as viscosity, spray angle, drug content per actuation, In-vitro drug diffusion study, Ex-vivo permeation study and In-vivo pharmacodynamics study by glucocorticoid induced model in rats. Pharmaco-tech. parameter results of optimized formulation showed satisfactory results. Comparative In-vitro drug permeation and ex-vivo permeation studies showed maximum drug release of 85% for optimized formulation incorporated with permeation enhance transcutol. In-vivo pharmacodynamic study showed better efficacy in treatment animals with optimized formulation and thus newly developed risedronate sodium sublingual spray proved to be effective used in treatment of osteoporosis.

In this study, a new self-microemulsifying drug delivery system (SMEDDS) based on pine essential oil has been designed and successfully developed to load cyclosporine A (CsA), a poorly water-soluble immunosuppressant, for oral CsA softgel manufacturing SMEDDS were prepared from pine essential oil and Capmul MCM as oily phases, Kolliphor RH 40 as surfactant, polyethylen glycol 400 as co-surfactant/co-solvent and none volatized co-solvent utilizing the pseudo-ternary phase diagrams. CsA SMEDDS loading 9.09% of CsA, dispersed in aqueous environments (water, pH 1.2, 4.5 and 6.8 buffers), presented an average droplet size of about 160 nm with polydispersity index (PdI) below 0.25, and were stable after 6 h of self-emulsification. Cytotoxicity of CsA SMEDDS on LLC-PK1 cells demonstrated that CsA SMEDDS had an IC50 value greater than that of the reference product, suggesting product safety against nephrotoxicity. CsA SMEDDS was filled into oval-5 soft gelatin capsules (softgels) containing 0.5% glycine which could avoid the crosslinking process. Addnl., CsA SMEDDS softgels presented the in vitro release profiles equivalent to that of Sandimmun Neoral at pH 1.2, 4.5 and 6.8. Therefore, the pine essential oil based SMEDDS was considered to be potential and suitable for developing softgels to enhance the oral bioavailability of CsA.

In this study, the volatile compound profiles of gurum seed oil were determined using two methods: supercritical CO2 extraction (SFE) and the screw press process (SPP). For volatile compounds extraction and identification, headspace solid-phase micro-extraction (HS-SPME) and GC-MS were used, resp. A total number of 56 volatile compounds were revealed and identified in oil extracted by SFE, while only 40 compounds were detected in extracted oil by SPP. Acids, aldehydes, esters, ketones, furans, and other components were present in the highest ratio in oil extracted by SFE. In contrast, alcs. and alkenes were found in the highest proportion in oil extracted by SPP. In this study, it was observed that SFE showed an increase in the amounts of volatile compounds and favorably impacted the aroma of gurum seed oil. The results reveal that different extraction methods significantly impact the volatile components of gurum seed oil, and this study can help evaluate the quality of the oil extracted from gurum seeds.

The purpose of this study was to develop and evaluate nanoemulsions (NEs) containing besifloxacin for ocular drug delivery. Pseudo ternary phase diagrams were constructed using Triacetin (oil), Cremophor RH 40 (surfactant), and TranscutolP (co-surfactant) to identify NE regions. Six formulations were developed by low-energy emulsification method and then evaluated for size, refractive index, pH, osmolality, viscosity, and drug release. After accelerated phys. stability and bovine conrneal permeation studies, NE2 was chosen as optimized formulation forantimicrobial efficacy, and hens egg test-chorioallantoic membrane (HET-CAM) tests. The particle size of optimum NE was 14 nm with a narrow size distribution. Moreover, other physicochem. characterizations were in the acceptable range for ocular administration. Besifloxacin-loaded NEs demonstrated sustained release pattern and 1.7-fold higher permeation compared with the control suspension in the ex vivo transcorneal permeation study. HET-CAM test indicated no irritation, and HL% revealed no damage to the tissue, so the optimum NE is well tolerated by the eye. In vitro antimicrobial evaluation, showed comparative efficacy of lower drug-loaded NE (0.2%) vs. 0.6% besifloxacin suspension (equal concentration to com. besifloxacin eye drop). In conclusion, besifloxacin-loaded NEs could be considered as a suitable alternative to the marketed suspension for treating bacterial eyeinfections.

Phase-separation absorbent has attracted much attention to reduce energy consumption for CO2 capture, storage and utilization. In this work, to improve handling of the absorbents, we have developed a new semi-solid absorbent, phase-separation gel, which became clouded gel by CO2 absorption and returned to transparent by CO2 desorption. We measured amounts of CO2 absorption at 50 C and desorption at 90 C with 10 vol% CO2 gas mixed with N2 gas. A cyclic capacity defined by the CO2 desorption amount of a phase-separation gel with solvent of 2-(ethylamino)ethanol/diethylene glycol di-Et ether was larger than those of the other homogeneous gels with 2-(ethylamino)ethanol/diethylene glycol monoethyl ether or diethylene glycol. Furthermore, the initial desorption flux of the phase-separation gel was larger than that of the absorbent solvent, suggesting that the phase-separation absorbent fixed inside the gels improves the initial desorption flux because of the larger interfacial area between the CO2-rich and CO2-lean phases inside the gel.
