Yu, Cheng-wei;Luo, Ting;Xie, Tian;Li, Jing;Deng, Ze-yuan research published 《 Classified processing of different rice bran fractions according to their component distributions》, the research content is summarized as follows. Rice grains were obtained from two Chinese provinces (Hubei and Guangxi) and then their rice bran was abraded into four equal fractions by weight from outside to inside. The component distributions in these four fractions were then investigated. The contents of total lipid-soluble antioxidant showed no significant difference between rice bran from the Hubei province and the Guangxi province. But the content of total water-soluble antioxidant was 1690.20 ± 86.40 mg kg-1 for rice bran from the Hubei province, significantly higher than those from the Guangxi province (1014.67 ± 71.09 mg kg-1). Obvious differences between four rice bran fractions or two provinces were proved by principal component anal. (PCA). Lipase activities decreased significantly from outer fractions to inner fractions, and lipids mainly distributed in three outer fractions. The essential element contents decreased significantly, but a main heavy metal element (Pb) in cereal increased significantly to 0.36 mg kg-1 from outer fractions to inner fractions, which was just slightly below the upper limit of Chinese Standard (0.40 mg kg-1). Although protein contents had no significant difference in four fractions, the amino acid compositions were quite different. In summary, different fractions of rice bran had different compositions, which could provide guidance for the classified processing of rice bran.

Yu, Changyue;Zhang, Baoli;Hu, Nan;Li, Lisong;Wang, Simiao;Zhang, Ziyuan;Wei, Tingyu;Zhao, Yingying;Wei, Hua;Hu, Qingbiao;Li, Yingdong research published 《 Comparative metabolomic analysis of Chinese mitten crab (Eriocheir sinensis) during the daily cycle》, the research content is summarized as follows. The circadian rhythm of aquatic animals has been a subject of research in a diverse range of studies. However, research on the circadian rhythm of the economically important crustacean Eriocheir sinensis is scarce although this species exhibits an obvious circadian rhythm. In this study, we applied liquid chromatog.-mass spectrometry-based metabolomics to identify key metabolites in the hepatopancreas of E. sinensis under the 24-h circadian rhythm. Through comparative anal. of the metabolites obtained at different time points (06:00 vs. 12:00 h, 06:00 vs. 18:00 h, and 06:00 vs. 24:00 h), 431 differentially expressed metabolites were identified and classified. The Kyoto Encyclopedia of Genes and Genomes pathway anal. showed that the differentially expressed metabolites were enriched in some circadian rhythm-related pathways, especially metabolic pathways related to nutrient digestion and absorption. Moreover, several pathways with high impact factors such as “Intestinal immune network for IgA production” were identified, and their functions were mostly related to immunity. This is the first study to identify the metabolites in the hepatopancreas of a crustacean through metabolomic anal. It provides novel insights into the circadian rhythm of E. sinensis, which could benefit its culture and further increase its economic value.

Fmoc-L-Ser-OH is a synthetic peptide that belongs to the group of glycopeptides. It is used as a model for such compounds and has been shown to have antimicrobial activity in vitro against gram-positive bacteria, especially Staphylococcus epidermidis. This compound was synthesized from 3-mercaptopropionic acid and chloride in the presence of hydroxyl groups and epidermal growth factor. Yin, Huawu;Zhou, Xiuman;Huang, Yen-Hua;King, Gordon J.;Collins, Brett M.;Gao, Yanfeng;Craik, David J.;Wang, Conan K. research published 《 Rational Design of Potent Peptide Inhibitors of the PD-1:PD-L1 Interaction for Cancer Immunotherapy》, the research content is summarized as follows. Peptides have potential to be developed into immune checkpoint inhibitors, but the target interfaces are difficult to inhibit. Here, we explored an approach to mimic the binding surface of PD-1 to design inhibitors. Mimicking native PD-1 resulted in a mimetic with no activity. However, mimicking an affinity-optimized PD-1 resulted in the peptide mimetic MOPD-1 that displayed nanomolar affinity to PD-L1 and could inhibit PD-1:PD-L1 interactions in both protein- and cell-based assays. Mutagenesis and structural characterization using NMR spectroscopy and X-ray crystallog. revealed that binding residues from the high affinity PD-1 are crucial for the bioactivity of MOPD-1. Furthermore, MOPD-1 was extremely stable in human serum and inhibited tumor growth in vivo, suggesting it has potential for use in cancer immunotherapy. The successful design of an inhibitor of PD-1:PD-L1 using the mimicry approach described herein illustrates the value of placing greater emphasis on optimizing the target interface before inhibitor design and is an approach that could have broader utility for the design of peptide inhibitors for other complex protein-protein interactions.

Here the authors report a new series of adenosine receptor A2A antagonists based on a 4-amino-5-carbonitrile pyrimidine template. Compounds from this new template exhibit excellent potency and ligand efficiency with low cytochrome P 450 inhibition. Although the clearance remains moderate to high, the leading compound, when dosed orally as low as 3 mg/kg, demonstrated excellent efficacy in the haloperidol induced catalepsy rat model for Parkinson’s disease.

Yang, Yurong;Zhong, Haiyan;Yang, Ning;Xu, Shuaizhe;Yang, Tao research published 《 Quality improvement of sweet rice wine fermented with Rhizopus delemar on key aroma compounds content, phenolic composition, and antioxidant capacity compared to Rhizopus oryzae》, the research content is summarized as follows. The pure culture fermentation has led to less flavorful rice wine and relatively lower bioactive substance level compared to traditional mixed culture fermentation; however, a pure strain is easily controlled by industrialized producers. The purpose of the present research was to screen a species of Rhizopus for improving the flavor deficiency and antioxidant function of sweet rice wine. Seven rice wine samples fermented with isolated strains were analyzed for their total phenolics, total flavonoids, peptide and antioxidant activity using spectrophotometry, as well as ethanol, Et acetate, β-phenethyl alc., and volatile alc. contents measured by headspace gas chromatog. (HS-GC), the further principal component anal. determined Rhizopus delemar rice wine better on aroma and antioxidant capacity. A comparison of phenolics profile between R. delemar and R. oryzae rice wines was made based on the measurement data of ultrahigh-performance liquid chromatog. coupled with Q-exactive orbitrap mass spectrometry (UHPLC-QE-MS) data. Thirty-two phenolics were identified in sweet rice wine. Overall, the results presented in this study showed that a strain of R. delemar is available for the improvement of flavor and antioxidant activity in sweet rice wine, which has the great potential to be applied to industrialized products.

The aim of this article was to design a self-emulsifying drug delivery system (SEDDS) of loaded cepharanthine (CEP) to improve the oral bioavailability in rats. Based on the solubility determination and pseudo-ternary phase diagram, iso-Pr palmitate (IPP) was chosen as the oil phase. Meanwhile, Cremophor RH40 and Macrogol 200 (PEG 200) were chosen as the emulsifier and co-emulsifier, resp. This prescription was further optimized by using central composite design of response surface methodol. The optimized condition was CEP:IPP:Cremophor RH40:PEG 200=3.6:30.0:55.3:11.1 in mass ratio with maximum drug loading (36.21 mg/mL) and the min. particle size (36.70 nm). The constructed CEP-SEDDS was characterized by dynamic light scattering, transmission electron microscopy, in vitro release and stability studies. The dissolution level of CEP-SEDDS was nearly 100% after 30 min in phosphate-buffered saline (PBS, pH 6.8) which was higher than that of the pure CEP (approx. 20%). In addition, in vivo pharmacokinetic study in rats showed that CEP-SEDDS dramatically improved bioavailability compared with active pharmaceutical ingredient (API) (the relative bioavailability was 203.46%). In this study, CEP-SEDDS was successfully prepared to enhance the oral bioavailability which might facilitate to increase its better clin. application.

Yang, Qi-Liang;Liu, Ying;Liang, Lei;Li, Zhi-Hao;Qu, Gui-Rong;Guo, Hai-Ming research published 《 Facilitating Rh-Catalyzed C-H Alkylation of (Hetero)arenes and 6-Arylpurine Nucleosides (Nucleotides) with Electrochemistry》, the research content is summarized as follows. An electrochem. approach to promote the ortho C-H alkylation of (hetero)arenes via rhodium catalysis under mild conditions was described. This approach features mild conditions with high levels of regio- and monoselectivity that tolerated a variety of aromatic and heteroaromatic groups and offered a widely applicable method for late-stage diversification of complex mol. architectures including tryptophan, estrone, diazepam, nucleosides and nucleotides. Alkyl boronic acids, esters and alkyl trifluoroborates were demonstrated as suitable coupling partners. The isolation of key rhodium intermediates and mechanistic studies provided strong support for a rhodium(III/IV or V) regime.
