The identification of novel inhaled p38α/β mitogen-activated protein kinases (MAPK) (MAPK14/11) inhibitors suitable for the treatment of pulmonary inflammatory conditions has been described. A rational drug design approach started from the identification of a novel tetrahydronaphthalene series, characterized by nanomolar inhibition of p38α with selectivity over p38γ and p38δ isoforms. SAR optimization of 1c is outlined, where improvements in potency against p38α and ligand-enzyme dissociation kinetics led to several compounds showing pronounced anti-inflammatory effects in vitro (inhibition of TNFα release). Targeting of the defined physicochem. properties allowed the identification of compounds 3h, 4e, and 4f, which showed, upon intratracheal instillation, low plasma levels, prolonged lung retention, and anti-inflammatory effects in a rat acute model of a bacterial endotoxin-induced pulmonary inflammation. Compound 4e (I), in particular, displayed remarkable efficacy and duration of action and was selected for progression in disease models of asthma and chronic obstructive pulmonary disease (COPD).

Arbour, Christine A.;Imperiali, Barbara research published 《 Backbone-anchoring, solid-phase synthesis strategy to access a library of peptidouridine-containing small molecules》, the research content is summarized as follows. Nucleoside diphosphate sugar (NDP-sugar) substrates provide the inspiration for nucleoside analog inhibitor scaffolds. By employing solid-phase synthesis, we provide a method to access a library of peptidouridine inhibitors with both minimal compound handling and purification steps. Specifically, this strategy is exemplified by generating uridine diphosphate sugar (UDP-sugar) mimics, which allow for compound elaboration by altering the dipeptide composition, the N-terminal linkage, and a pendant aryl group. To exemplify the versatility, 41 unique nucleoside analogs are presented.

Araujo, Gustavo Parreira;Martins, Felipe Terra;Taveira, Stephania Fleury;Cunha-Filho, Marcilio;Marreto, Ricardo Neves research published 《 Effects of Formulation and Manufacturing Process on Drug Release from Solid Self-emulsifying Drug Delivery Systems Prepared by High Shear Mixing》, the research content is summarized as follows. This study sought to investigate the influence of formulation and process factors of the high shear mixing (HSM) on the properties of solid self-emulsifying drug delivery systems (S-SEDDS) containing the model drug carvedilol (CAR). Firstly, liquid SEDDS (L-SEDDS) were prepared by mixing castor oil with different proportions of surfactant (Solutol or Kolliphor RH40) and cosolvent (Transcutol or PEG400). A miscible L-SEDDS with high drug solubility (124.3 mg/g) was selected and gave rise to 10% (m/m) CAR loaded-emulsion with reduced particle size. Then, a factorial exptl. design involving five component′s concentration and two process factors was used to study the solidification of the selected L-SEDDS by HSM. CAR content, diffractometric profile, and in vitro dissolution were determined Morphol. and flow analyses were also performed. Porous and spherical particles with mean sizes ranging from 160 to 210 μm were obtained. Particle size was not affected by any formulation factor studied. Powder flowability, in turn, was influenced by L-SEDDS and crospovidone concentration CAR in vitro dissolution from S-SEDDS was significantly increased compared to the drug as supplied and was equal (pH 1.2) or lower (pH 6.8) than that determined for L-SEDDS. Colloidal silicon dioxide decreased drug dissolution, whereas an increase in water-soluble diluent lactose and L-SEDDS concentration increased CAR dissolution The proper selection of liquid and solid constituents proved to be crucial to developing an S-SEDDS by HSM. Indeed, the results obtained here using exptl. design contribute to the production of S-SEDDS using an industrially viable process.

In the current study, a novel self-nanoemulsifying drug delivery system (SNEDDS) was developed in order to improve the oral bioavailability of delafloxacin (DLF). Various oils, surfactants and co-surfactants were screened by solubility studies. Based on solubilization, potential of DLF in various vehicles, Lauroglycol-90 (oil), Tween 80 (surfactant) and Transcutol-HP (co-surfactant) were selected for the development of DLF loaded SNEDDS (DLF-SNEDDS). Four formulas of DLF loaded SNEDDS (DLF1-DLF4) were prepared and evaluated for their droplet size, polydispersity index (PDI), zeta potential (ZP), refractive index (RI), percent transmittance (%T), thermodn. stability, emulsifying efficiency and in vitro release studies. Among developed DLF loaded SNEDDS (DLF1-DLF4), DLFI was found optimum with a droplet size (73.3 ± 6.5 nm) PDI (0.298) ZP (-22.3) RI (1.334 ± 0.05) and %T (99.2 ± 0.09). Optimized DLF loaded SNEDDS (DLF1) demonstrated a superior anti-microbial activity against tested gram pos. and gram neg. strains with no resistance. The pharmacokinetic studies showed that oral bioavailability of DLF was significantly improved (3- fold) by optimized DLF loaded SNEDDS (DLF1) in comparison to pure DLF suspension. Hence, the results of this work suggest that the developed SNEDDS could be a potential carrier in enhancing bioavailability and therapeutic efficacy of DLF.

It also decreases levels of adhesion molecules and downregulates inflammatory response genes. Sinapinic acid has been shown to reduce inflammation by inhibiting the formation of proinflammatory mediators, such as prostaglandin E2 or leukotriene B4, in endothelial cells and mammary epithelial cells.
Sinapic acid is a phenylpropanoid hydroxycinnamic acid with diverse biological activities. Sinapic acid inhibits collagen-induced human platelet aggregation by up to 70% in vitro (IC50 = 1.03 mM). It scavenges 2,2-diphenyl-1-picrylhydrazyl (DPPH; ) and 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonate) (ABTS) free radicals with IC50 values of 8.3 and 5.4 μg/ml, respectively. Cellat, Mustafa;Isler, Cafer Tayer;Uyar, Ahmet;Kuzu, Muesluem;Aydin, Tuba;Etyemez, Muhammed;Tuerk, Erdinc;Yavas, Ilker;Guevenc, Mehmet research published 《 Protective effect of Smilax excelsa L. pretreatment via antioxidant, anti-inflammatory effects, and activation of Nrf-2/ HO -1 pathway in testicular torsion model》, the research content is summarized as follows. The protective effects of the ethanol extract of Smilax excelsa L. (SE) leaves were investigated on testicular tissue of rats with a torsion model in this study. The chem. composition of the extract was detected by means of liquid chromatog. with tandem mass spectrometry (LC-MS/MS). SE extract was given for 21 days before torsion was created in the treatment group. The sperm parameters of the torsion group were impaired, and there was an increase in MDA level as well as a decrease in GSH level and GPx activity compared to the control group. TNF-α and NF-κB levels in the torsion group increased as compared to those in the control group. The expression levels of Nrf-2 and HO-1 were lower in the torsion group than those in the control group. The SE pretreatment group has improved sperm, oxidative stress, and inflammatory markers when compared to the torsion group, and the Nrf-2/HO-1 pathway was activated. Practical applications : Smilax excelsa L. is a plant with economic value used in traditional medicine in the treatment of stomachache, bloating, and breast cancer in Northwest Anatolia. It has an antioxidant effect due to the flavonoids and anthocyanins it contains. The protective effect against ischemia-reperfusion-induced tissue and reproductive damage in testicular tissue were demonstrated with the study. When the histol. examinations of the tissues were evaluated, it was found that morphol. structure of the tissues was retained in the treatment group. The findings indicate that SE prevents tissue damage in the torsion model by antioxidant and anti-inflammatory effects and activating Nrf-2/HO-1 pathway.

R.;Barros, Lillian research published 《 Sequential steps of the incorporation of bioactive plant extracts from wild Italian Plantago coronopus L. and Cichorium intybus L. leaves in fresh egg pasta》, the research content is summarized as follows. The application of bioactive extracts from Cichorium intybus L. and Plantago coronopus L. species were incorporated as a functional ingredient in fresh egg pasta (Fettuccine). In that sense, a pasta making procedure was accessed using different concentrations of the plant extracts (0.25-0.63 mg/g), drying times (20-420 min) and drying temperatures (40-90 °C; only for P. coronopus enriched pasta), to screen an optimal factor selection in the pasta making procedure and to enhance the bioactive properties of the final product. In the chem. characterization of the plant extracts, twenty-five phenolic compounds were tentatively identified (twenty compounds belonging to phenolic acid and phenylpropanoid classes and five belonging to the flavonoid sub-class) and a strong synergy between the plant extract concentration and the drying time was showed. The analyzed antioxidant properties were enhanced by the phenolic compounds of the extracts and a new functional food with higher bioactive quality was developed.

530-59-6, Sinapinic acid is a chemical compound that is the dihydroxybenzoic acid derivative of sinapic acid. It has been shown to have anti-inflammatory properties in vitro and in vivo. Sinapinic acid inhibits the activity of various enzymes, such as cyclooxygenase (COX), lipoxygenase (LOX), and 5-lipoxygenase-activating protein (FLAP).
Sinapic acid (200 μM) reduces colony formation of SW480 human colon carcinoma cells by 4-fold. It also inhibits colony formation of E. coli, S. enteritidis, and S. aureus on agar (MICs = 2.2, 2, and 1.8 mM, respectively). In vivo, sinapic acid (4 mg/kg, p.o.) increases the time spent in the open arms of the elevated plus maze by approximately 15% in mice, an effect that can be blocked by the GABAA receptor antagonists flumazenil and bicuculline. Sinapic acid is also commonly used as a matrix in protein mass spectrometry.
Sinapic acid analytical standard provided with w/w absolute assay, to be used for quantitative titration.
Sinapic acid is an hydroxycinnamic acid derivative that occurs naturally in Brassicaceae species.
cis-Sinapic acid, also known as cis-sinapate or synapitic acid, belongs to the class of organic compounds known as hydroxycinnamic acids. Hydroxycinnamic acids are compounds containing an cinnamic acid where the benzene ring is hydroxylated. cis-Sinapic acid is considered to be a practically insoluble (in water) and relatively neutral molecule. Within the cell, cis-sinapic acid is primarily located in the cytoplasm. Outside of the human body, cis-sinapic acid can be found in common pea and pulses. This makes cis-sinapic acid a potential biomarker for the consumption of these food products.
Cis-sinapic acid is a 3-(4-hydroxy-3,5-dimethoxyphenyl)prop-2-enoic acid in which the double bond has cis-configuration. It has been isolated from the shoots of alfalfa. It has a role as a plant metabolite., Computed Properties of 530-59-6

Ethers lack the hydroxyl groups of alcohols. Without the strongly polarized O―H bond, ether molecules cannot engage in hydrogen bonding with each other. 530-59-6, formula is C11H12O5, Name is 3,5-Dimethoxy-4-hydroxycinnamic acid. Ethers do have nonbonding electron pairs on their oxygen atoms, however, and they can form hydrogen bonds with other molecules (alcohols, amines, etc.) that have O―H or N―H bonds. Mesquite (Prosopis juliflora) is a pod found in the semiarid Northeast region of Brazil. It is widely used as livestock feed and its byproduct (the grain) has potential for human food. Thus, this article aims was to obtain flour from mesquite grains under different drying temperatures (50, 60, and 70°C) and classify them by their phys.-chem., microstructural, technol., antioxidant, bioactive, and pasting properties. Both the mesquite grain and the mesquite grain flour showed a high amount of proteins and low-fat content. Analyses on morphol. and viscosity indicated the presence of starch. Among the temperatures studied, the flour dried at 60°C presented the best results for the capacity of water and oil absorption, antioxidant activities, and total phenolic compounds Seventeen compounds were detected by chromatog., with kaempferol, catechin, and quinine being the most abundant. This confirms the potential of mesquite flour as a relevant source of proteins and phenolic compounds for food products.

D.;Clemens, Daniel;Kent, Ben;Lancellotti, Marcelo;de Araujo, Daniele R.;de Paula, Eneida research published 《 A pre-formulation study of tetracaine loaded in optimized nanostructured lipid carriers》, the research content is summarized as follows. Tetracaine (TTC) is a local anesthetic broadly used for topical and spinal blockade, despite its systemic toxicity. Encapsulation in nanostructured lipid carriers (NLC) may prolong TTC delivery at the site of injection, reducing such toxicity. This work reports the development of NLC loading 4% TTC. Structural properties and encapsulation efficiency (%EE > 63%) guided the selection of three pre-formulations of different lipid composition, through a 23 factorial design of experiments (DOE). DLS and TEM analyses revealed average sizes (193-220 nm), polydispersity (< 0.2), ζ potential |- 21.8 to – 30.1 mV| and spherical shape of the nanoparticles, while FTIR-ATR, NTA, DSC, XRD and SANS provided details on their structure and physicochem. stability over time. Interestingly, one optimized pre-formulation (CP-TRANS/TTC) showed phase-separation after 4 mo, as predicted by Raman imaging that detected lack of miscibility between its solid (cetyl palmitate) and liquid (Transcutol) lipids. SANS analyses identified lamellar arrangements inside such nanoparticles, the thickness of the lamellae been decreased by TTC. As a result of this combined approach (DOE and biophys. techniques) two optimized pre-formulations were rationally selected, both with great potential as drug delivery systems, extending the release of the anesthetic (> 48 h) and reducing TTC cytotoxicity against Balb/c 3T3 cells.
